Co-transformation of a tropical maize endophytic isolate of Fusarium verticillioides (synonym F. moniliforme) with gusA and nia genes Genet. Mol. Biol.

A tropical endophytic isolate of the fungus Fusarium verticillioides (synonym Fusarium moniliforme) obtained from Zea mays was co-transformed with plasmid pNH24 containing the Fusarium oxysporum nitrate reductase nia gene and plasmid pNOM 102 carrying the Escherichia coli b-glucuronidase gusA gene. Transformation frequency for the nia marker was 75 transformants mg-1 vector DNA and introduction of the gusA gene by co-transformation was 57.2% as indicated by the presence of the GUS+ phenotype on plates. ABSTRACT: Gene stacking refers to the introduction of two or more transgenes of agronomic interest in the same plant. The main methods for genetically engineering plants with gene stacking involve (i) the simultaneous introduction, by the co-transformation process, and (ii) the sequential introduction of genes using the re-transformation processes or the sexual crossing between separate transgenic events. In general, the choice of the best method varies according to the species of interest and the availability of genetic constructions and preexisting transgenic events. Transformation of an Argentine spring wheat genotype: optimization of the protocols for particle bombardment of excised immature embryos and rapid isolation of transgenic plants JBAG

Klein Brujo was found to be a promising Argentine spring wheat genotype for transformation studies. In the present work we optimized the biolistic transformation of embryogenic scutellar calli from this genotype. We first identified scutellar callus induction media (SCIM) most promising for in vitro embryogenic plant regeneration with Klein Brujo. We then co-bombarded embryos of Klein Brujo and, for comparison, of Bobwhite, a highly transformable wheat line, on these media with 1 μm gold particles coated with two plasmids.
